在0℃下,將異喹啉(13a)(1.83 mL, 15.50 mmol)緩慢滴加至濃硫酸(17 mL)中,同時保持?jǐn)嚢?。將反?yīng)混合物冷卻至-25℃,隨后分批加入硼氫化鈉(6.34 g, 35.65 mmol),控制加入速率以維持反應(yīng)溫度在-25℃至-20℃之間。在此溫度下持續(xù)反應(yīng)1小時后,允許反應(yīng)混合物自然升溫至室溫。將反應(yīng)混合物小心倒入碎冰中,并用濃氨水溶液調(diào)節(jié)pH至7.0。所得漿液在0℃下攪拌1小時,隨后過濾,并用冰水洗滌。粗產(chǎn)物經(jīng)風(fēng)干后,通過柱色譜法純化,得到5,8-二溴異喹啉(13b)(2.5 g, 56%)。質(zhì)譜(EI) m/z: 286([M+1]+)。1H NMR (400 MHz, DMSO-d6) δ: 9.48 (s, 1H), 8.78 (d, J = 6 Hz, 1H), 8.06 (d, J = 8 Hz, 1H), 7.98 (dd, J = 6 Hz, J = 8 Hz, 2H)。
