在氬氣保護下,將二氟甲基苯基砜(192 mg,1 mmol)和元素碘(508 mg,4 mmol)溶解于DMF(4 mL)中,置于Schlenk燒瓶內。將體系冷卻至-30℃,通過注射器緩慢滴加t-BuOK(448 mg,4 mmol)的DMF(4 mL)溶液。保持反應溫度在-30℃至-20℃之間,持續(xù)攪拌1小時,反應進程通過19F NMR跟蹤監(jiān)測。反應完成后,于-50℃下加入1N HCl水溶液(5 mL)淬滅反應,隨后緩慢升溫至室溫。加入飽和NaCl水溶液(10 mL),用乙醚(15 mL×3)萃取反應混合物。合并有機相,用無水MgSO4干燥,過濾后減壓濃縮除去溶劑。殘余物通過硅膠柱色譜法(洗脫劑:己烷/乙酸乙酯=9:1)純化,得到目標產(chǎn)物二氟碘甲基苯基砜(294 mg,收率92%),為無色固體,遇光易變紅。產(chǎn)物表征數(shù)據(jù)如下:1H NMR (500 MHz, CDCl3): δ 7.63 (t, J = 7.4 Hz, 2H), 7.79 (t, J = 7.4 Hz, 1H), 7.99 (d, J = 7.5 Hz, 2H); 13C NMR (125 MHz, CDCl3): δ 102.5 (t, J = 355 Hz), 128.1, 129.7, 131.3, 136.1; 19F NMR (470 MHz, CDCl3): δ -52.2; MS (EI, m/z): 318 (M+), 177, 142, 127; HRMS (EI): m/z calcd for C7H5F2IO2S (M+) 317.9023, found 317.9013.
