步驟I:將硼氫化鈉(5.20 g,0.137 mol)分批加入至4-溴-2-噻吩甲醛(25.0 g,0.131 mol)的無(wú)水四氫呋喃(400 mL)溶液中。反應(yīng)混合物在室溫下攪拌1.5小時(shí)。反應(yīng)完成后,緩慢加入飽和氯化銨溶液(100 mL)以淬滅反應(yīng)。隨后,用乙酸乙酯萃取反應(yīng)混合物,合并有機(jī)相并用飽和鹽水洗滌。有機(jī)層經(jīng)無(wú)水硫酸鈉干燥后,減壓濃縮,得到(4-溴-2-噻吩基)甲醇(25.02 g,收率99%),產(chǎn)物無(wú)需進(jìn)一步純化即可用于后續(xù)反應(yīng)。核磁共振氫譜(400 MHz, CDCl3): δ = 1.93(寬單峰,1H),4.79(單峰,2H),6.93(單峰,1H),7.18(雙峰,J = 1.5 Hz,1H)。
