以2,6-二溴-3-羥基吡啶(12g,47mmol)為起始原料,與碳酸鉀(6.0g,43mmol)、碘甲烷(10.1mL,162mmol)在DMSO(20mL)中混合形成懸浮液。將反應(yīng)混合物加熱回流2小時(shí)。反應(yīng)完成后,將混合物倒入水(60mL)中,于50℃溫和加熱并攪拌30分鐘。隨后,將反應(yīng)混合物冷卻至室溫,通過過濾收集沉淀的固體,并在真空條件下干燥。最后,將干燥后的殘余物從環(huán)己烷中結(jié)晶,得到目標(biāo)產(chǎn)物2,6-二溴-3-甲氧基吡啶(10.1g,產(chǎn)率80%),為淺橙色固體。產(chǎn)物經(jīng)1H NMR(400MHz,DMSO-d6)確認(rèn):δ= 7.66(d,J = 8.0Hz,1H),7.52(d,J = 8.0Hz,1H),3.90(s,3H);質(zhì)譜分析顯示ES + ve m/z 266,268,270(M + H)+。
