按照Yang等人所述的方法(參見(jiàn)J. Am. Chem. Soc., 1996, 118:9557),將中間體62研磨后,分次加入裝有15 mL濃硫酸的50 mL錐形瓶中,同時(shí)保持?jǐn)嚢璨囟染S持在90℃。緩慢加入乙酰胺,確保反應(yīng)混合物的溫度不超過(guò)105℃。加畢,將紫黑色溶液在90℃下繼續(xù)攪拌15分鐘,隨后冷卻至60℃,并倒入含有15克碎冰的燒杯中。繼續(xù)添加冰直至燒杯外壁觸感冰冷。通過(guò)過(guò)濾收集橙棕色沉淀,并在真空條件下干燥過(guò)夜,得到純度足以用于下一步驟的二氫吲哚-2,3-二酮(中間體63,0.77 g,收率27%)。中間體63亦可經(jīng)乙醇重結(jié)晶,獲得純橙色紅色針狀產(chǎn)物。1H NMR(400 MHz, DMSO-d6)δ 1.14(t, J = 7.5 Hz, 3H),2.56(q, J = 7.6 Hz, 2H),7.03(t, J = 7.5 Hz, 1H),7.35(d, J = 7.3 Hz, 1H),7.46(d, J = 7.6 Hz, 1H),11.11(s, 1H)。
