以2,4-二氧代-5-甲基-己酸乙酯為原料合成5-異丙基吡唑-3-甲酸乙酯的一般步驟:將2,4-二氧代-5-甲基-己酸乙酯(1g,5.37mmol)和水合肼(10.7mmol,9.64mol/L)在乙醇(20mL)中的混合物在75℃下,氮?dú)夥諊袛嚢?小時(shí)。反應(yīng)完成后,將混合物在真空下濃縮。殘余物通過(guò)硅膠柱色譜法純化,使用石油醚:乙酸乙酯(體積比2:1)作為洗脫劑,得到淺黃色固體5-異丙基吡唑-3-甲酸乙酯(0.55g,產(chǎn)率56%)。質(zhì)譜(ESI,正離子模式)m/z:183.1 [M + H]+。1H NMR(600MHz,CDCl3)δ:11.37(br s,1H),6.63(s,1H),4.40-4.37(m,2H),3.07-3.05(m,1H),1.43-1.40(m,3H),1.39-1.30(m,6H)。
