步驟1. 在2L三頸燒瓶中依次加入4-氯-2-(甲硫基)嘧啶-5-羧酸乙酯(100g, 0.43mol)、四氫呋喃(THF, 500mL)和三乙胺(TEA, 186mL, 1.29mol)。在攪拌下分批加入28%氫氧化銨水溶液(400mL),控制反應(yīng)溫度低于30℃。反應(yīng)2小時后,加入水(1000mL),通過真空蒸餾除去THF。將反應(yīng)混合物過濾,收集固體產(chǎn)物,并在50℃下真空干燥,得到白色固體4-氨基-2-(甲硫基)嘧啶-5-羧酸乙酯(90g, 收率99%)。產(chǎn)物經(jīng)LC/MS分析顯示m/z = 213.9 [M+H]+;1H-NMR (DMSO-d6, 400MHz) δ 8.57 (s, 1H), 8.03 (br, 1H), 7.65 (br, 1H), 4.27 (q, J = 7.5Hz, 2H), 2.46 (s, 3H), 1.29 (t, J = 7.2Hz, 3H)。
