以8-芐基-8-氮雜雙環(huán)[3.2.1]辛烷-3-酮肟為原料合成外向-8-芐基-8-氮雜雙環(huán)[3.2.1]辛-3-胺的一般步驟:[105] 在120℃下,將8-芐基-8-氮雜雙環(huán)[3.2.1]辛烷-3-酮肟(7.65g,33mmol)溶于1-戊醇(130mL)中。在2小時內(nèi)分批加入金屬鈉(7.6g,0.33mol,10.0當(dāng)量)。將混合物在回流條件下攪拌并加熱5小時,然后冷卻至5℃。緩慢用6M HCl酸化反應(yīng)混合物至pH值達(dá)到酸性,然后用6M HCl(3×100mL)進(jìn)一步萃取水相。通過加入5M NaOH將水層逐漸堿化至pH10。用EtOAc(3×100mL)萃取所得水溶液。合并有機(jī)萃取液,用MgSO4干燥,過濾,真空濃縮。粗產(chǎn)物通過柱色譜法(5-10%MeOH/CH2Cl2)純化,得到純的外向-8-芐基-8-氮雜雙環(huán)[3.2.1]辛-3-胺,為固體(2.60g,36%)。
