b)4-羥基-1-甲基吲哚的合成:在加氫裝置中,將4-(芐氧基)-1-甲基-1H-吲哚(0.250 g,1.12 mmol)溶于甲醇(2.25 mL)中,形成棕色溶液。向該溶液中加入5% Pd/C催化劑(0.119 g,1.12 mmol,預(yù)先在烘箱中干燥)。將混合物連接至氫化裝置,進(jìn)行三次脫氣操作后,充入氫氣。在最后一次脫氣后,于40 psi氫氣壓力下振蕩反應(yīng)16小時(shí)。反應(yīng)完成后,用甲醇(15 mL)稀釋反應(yīng)混合物,通過(guò)硅藻土層(2.5英寸,流速2 L/h)過(guò)濾,并用溫?zé)岬募状迹?5 mL)洗滌。濃縮有機(jī)濾液,得到0.160 g(產(chǎn)率97%)的黑色油狀產(chǎn)物。產(chǎn)物經(jīng)1H NMR(CDCl3)表征:δ 7.07(t,J = 8.24 Hz,1H),6.92(m,J = 8.24 Hz,2H),6.52(m,J = 2.47 Hz,2H),4.60(br s,1H),3.75(s,3H)。
