1. 將6-喹啉甲酸(9.50g,54.9mmol)溶于乙醇(250ml)中,加入濃硫酸(2ml),回流反應(yīng)8小時(shí)。反應(yīng)完成后,蒸發(fā)除去溶劑。將殘余物溶解于水中,用氫氧化鉀水溶液調(diào)節(jié)pH至8,過濾收集產(chǎn)物,真空干燥,得到6-喹啉甲酸乙酯(9.85g,收率89%),為淺棕色固體。熔點(diǎn):66-67℃。TLC(展開劑:CH2Cl2/MeOH/AcOH=9:0.5:0.1):Rf=0.52。 2. 將6-喹啉甲酸乙酯(9.80g,48.7mmol)溶于適量溶劑中,用1N HCl水溶液調(diào)節(jié)pH至2。加入20% Pd-Mohr催化劑(1.96g),在60℃、3巴氫氣壓力下氫化17小時(shí)。反應(yīng)完成后,反應(yīng)混合物通過硅藻土過濾。蒸發(fā)濾液,殘余物溶于乙酸乙酯和水中,用1N氫氧化鉀水溶液調(diào)節(jié)pH至16。分離有機(jī)相和水相,有機(jī)相用鹽水洗滌,無水Na2SO4干燥,蒸發(fā)溶劑,得到1,2,3,4-四氫-6-喹啉甲酸乙酯(8.72g,收率87%),為淺棕色固體。熔點(diǎn):68-70℃。GC-MS:[M+]=205。
