在氬氣保護(hù)下,將Pd(dba)2(275 mg,0.30 mmol)和三環(huán)己基膦(504 mg,1.80 mmol)的二惡烷溶液(30 mL)加入到攪拌中的5-溴吲哚-2-甲酸乙酯(6.0 g,22.4 mmol)混合物中。隨后加入KOAc(3.3 g,33.6 mmol)、雙(頻哪醇合)二硼(6.3 g,24.6 mmol)以及額外的二惡烷(20 mL)。將反應(yīng)混合物在80℃下攪拌3小時(shí),之后冷卻至室溫并通過硅藻土過濾。用EtOAc洗滌固體,合并濾液并濃縮,最后通過柱色譜法純化,得到目標(biāo)產(chǎn)物2-乙氧基羰基吲哚-5-硼酸頻哪醇酯(6.8 g,收率97%)。
