以3-氧代-4-(2,2,2-三氟乙酰基)哌啶-1-羧酸叔丁酯為起始原料,合成3-(三氟甲基)-4,5-二氫-1H-吡唑并[3,4-c]吡啶-6(7H)-羧酸叔丁酯的一般步驟如下:向1-叔丁氧基羰基-4-(三氟乙酰基)-3-哌啶酮(23.6g,0.08mol)的乙醇(250ml)溶液中加入一水合肼(4.16ml,0.086mol)。將反應(yīng)混合物在50℃下攪拌24小時(shí)。反應(yīng)完成后,將混合物冷卻至室溫,隨后進(jìn)行減壓濃縮。將濃縮后的殘余物在乙酸乙酯和飽和鹽水之間分配。分離有機(jī)層,用無水硫酸鈉干燥,過濾后減壓濃縮。所得殘余物通過乙酸乙酯/正戊烷混合溶劑重結(jié)晶,得到目標(biāo)化合物3-(三氟甲基)-4,5-二氫-1H-吡唑并[3,4-c]吡啶-6(7H)-羧酸叔丁酯,為淺黃色固體(6.5g,收率28%)。產(chǎn)物經(jīng)1H-NMR(400MHz,CDCl3)確認(rèn):δ 11.13(1H,寬單峰),4.40(2H,多重峰),3.66(2H,多重峰),2.70(2H,多重峰),1.48(9H,單峰);LC/MS分析顯示保留時(shí)間為2.90分鐘,質(zhì)譜(ES-)m/z 290([M-H]-,計(jì)算值C12H16F3N3O2的分子量為291)。
