a)4-溴-1-乙基-1H-吡唑的合成:在4-溴-1H-吡唑(5g,34mmol)的DMF溶液中加入碳酸鉀(K2CO3,11.75g,85.03mmol,2.5當(dāng)量)和碘乙烷(8g,51mmol,1.5當(dāng)量)。將反應(yīng)混合物在室溫下攪拌12小時(shí)。反應(yīng)完成后,按照中間體實(shí)施例5(c)的方法進(jìn)行驟冷和萃取操作。通過(guò)蒸餾去除溶劑,得到的粗產(chǎn)物采用柱色譜法(60-120目硅膠,40%乙酸乙酯的己烷溶液作為洗脫劑)進(jìn)行純化,最終得到目標(biāo)產(chǎn)物1-乙基-4-溴吡唑,收率為84%(5g)。產(chǎn)物結(jié)構(gòu)通過(guò)1H NMR(300MHz,DMSO-d6)和LC-MS(ESI)進(jìn)行確認(rèn):1H NMR (300MHz, DMSO-d6) δ 8.02 (s, 1H), 7.55 (s, 1H), 4.15 (q, 2H), 1.37 (t, 3H); LC-MS (ESI): 計(jì)算質(zhì)量:175.03; 實(shí)測(cè)質(zhì)量:177.0 [M + H]+(保留時(shí)間:0.56min)。
