在-5℃條件下,向26.50g(0.14mol)2-溴苯甲酸乙酯(4a)溶于35mL無水二乙醚的溶液中,緩慢加入由9.1g(0.37mol)鎂和53.3g(0.37mol)新蒸餾的甲基碘在85mL無水乙醚中制備的甲基碘化鎂格氏試劑。反應(yīng)完成后,加入38%氯化銨溶液(58mL)淬滅反應(yīng)。通過傾析分離醚層,并用乙醚(3×35mL)萃取剩余的糊狀沉淀。合并所有醚層萃取物,用無水硫酸鈉干燥,隨后蒸發(fā)溶劑。得到產(chǎn)物2-(2-溴苯基)-2-丙醇19.40g,收率82%,純度95%(GLC測定)。產(chǎn)物物理性質(zhì):沸點(diǎn)99-100℃(5mmHg),d420=1.1576,nD20=1.5420;MRD實(shí)測值46.43,計(jì)算值46.55。文獻(xiàn)數(shù)據(jù)對比:熔點(diǎn)23.7℃,沸點(diǎn)79.2℃(2.2mmHg),nD20=1.5416。IR光譜特征吸收峰(ν, cm-1):3460, 1175(OH),1370, 1385 [C(CH3)2],690(C-Cl)。1H NMR光譜數(shù)據(jù)(δ, ppm):1.64s(6H, CH3),2.46s(1H, OH),6.9-7.7m(4H, Harom)。元素分析實(shí)測值(%):C 63.00, H 6.42, Cl 20.71;分子式C9H11ClO的計(jì)算值(%):C 63.35, H 6.50, Cl 20.78。
