通用程序:在1小時(shí)內(nèi),將乙酸(AcOH,100%,140 mL,2.44 mol)緩慢滴加到含有在0-5°C下攪拌的氫化鈉(NaBH4,20.0 g,0.53 mol)和氯仿(CHCl3,220 mL)的燒瓶中。將所得混合物在0-5°C下繼續(xù)攪拌1.5小時(shí)。隨后,將N-甲基哌嗪(28.0 mL,0.25 mol)和對(duì)硝基苯甲醛(43.4 g,0.26 mol)溶于氯仿(60 mL)的溶液加入上述混合物中。將反應(yīng)混合物在0-5°C下攪拌1小時(shí),然后在室溫下攪拌12小時(shí)。反應(yīng)完成后,用蒸餾水(150 mL)和碳酸鈉(Na2CO3)處理混合物,調(diào)節(jié)pH至8.0-9.0。水相用乙酸乙酯(EtOAc,2×100 mL)萃取,合并有機(jī)層,用蒸餾水(1×100 mL)洗滌,無(wú)水硫酸鈉(Na2SO4)干燥。過濾并蒸發(fā)溶劑,得到1-甲基-4-(4-硝基苯)哌嗪(4a),為淡黃色油狀物;產(chǎn)量:61.6 g,收率99%。
