如方案9的步驟9-i所示,將4-羥基-2-甲基-6-氧代-1,6-二氫吡啶-3-羧酸乙酯(Accla Biochem Inc.)懸浮于50 mL的三氯氧磷(POCl3)中,在氮?dú)獗Wo(hù)下加熱至90℃,反應(yīng)5小時(shí)。反應(yīng)完成后,冷卻反應(yīng)混合物,并在減壓條件下濃縮。隨后,在攪拌下向深色油狀物中加入冰,接著加入乙酸乙酯和水進(jìn)行萃取。有機(jī)相依次用水和飽和食鹽水洗滌,經(jīng)無(wú)水硫酸鈉干燥。過(guò)濾后,減壓除去溶劑,粗產(chǎn)物通過(guò)硅膠柱色譜法純化,得到2-甲基-4,6-二氯煙酸乙酯(化合物2030),為淺黃色油狀物,產(chǎn)率62%。質(zhì)譜(ESMS)顯示分子離子峰(M + H)為234/236/238;1H NMR(CDCl3)δ 7.27(s, 1H),4.40(q, 2H),2.55(s, 3H),1.41(t, 3H)。
