一般步驟:向5-碘-2'-脫氧胞苷(2.2 g,6.23 mmol)的無水DMF(130 mL)溶液中加入咪唑(467 mg,6.85 mmol)。將反應(yīng)混合物冷卻至0℃,并在5分鐘內(nèi)緩慢加入叔丁基二甲基甲硅烷基氯(TBDMSCl)(1.33 g,6.85 mmol)。反應(yīng)混合物在室溫下攪拌18小時后,減壓濃縮以去除揮發(fā)物。殘余物通過硅膠快速柱色譜法純化,洗脫劑為EtOAc:MeOH(95:5至90:10),得到目標(biāo)產(chǎn)物4-氨基-1-((2R,4S,5R)-5-(((叔丁基二甲基硅烷基)氧基)甲基)-4-羥基四氫呋喃-2-基)-5-碘嘧啶-2(1H)-酮(2.10 g,72%收率)及未反應(yīng)的原料(490 mg)。產(chǎn)物表征數(shù)據(jù)如下:1H NMR (d6-DMSO) δ 0.11 (s, 3H, CH3), 0.12 (s, 3H, CH3), 0.89 (s, 9H, 3×CH3), 1.90 (ddd, J = 13.2, 7.7, 5.7 Hz, 1H, H-2'), 2.18 (ddd, J = 13.2, 5.7, 2.3 Hz, 1H, H-2'), 3.72 (dd, J = 11.5, 3.6 Hz, 1H, H-5'), 3.80 (dd, J = 11.5, 2.8 Hz, 1H, H-5'), 3.86-3.89 (m, 1H, H-4'), 4.14-4.18 (m, 1H, H-3'), 5.22 (d, J = 4.1 Hz, 1H, OH), 6.09 (dd, J = 7.8, 5.8 Hz, 1H, H-1'), 6.60 (br s, 1H, NH), 7.81 (br s, 1H, NH), 7.94 (s, 1H, H-6); MS (ES): m/z (百分比) (M + H)+ 468 (90%).
