3-碘吲唑的合成按照Bocchi先前報(bào)道的方法[28]進(jìn)行,并稍作修改。具體步驟如下:將1H-吲唑(3g,25.4mmol)、碘(12.7g,50.03mmol)和氫氧化鉀(5.34g,95.25mmol)溶解于DMF(7mL)中,室溫下攪拌反應(yīng)3小時(shí)。反應(yīng)完成后,加入飽和亞硫酸氫鈉溶液(150mL)淬滅反應(yīng),此時(shí)有沉淀生成。通過真空過濾收集沉淀,并用蒸餾水(3×30mL)洗滌。所得固體在30℃真空烘箱中干燥過夜,最終獲得6.17g淺黃色固體產(chǎn)物,收率為100%。產(chǎn)物表征數(shù)據(jù)如下:熔點(diǎn)136-138℃(文獻(xiàn)值[36]:134-136℃);IR(KBr)ν(cm-1):3086(NH),424(C-I);1H-NMR δ(ppm):13.50(1H,s,H-1),7.55(1H,d,J=8.6Hz,H-7),7.45-7.40(2H,m,H-6和H-4),7.19(1H,dd,J=7.5Hz,H-5);13C-NMR δ(ppm):140.41,127.22,126.79,121.23,120.39,110.51,93.49;HRMS(計(jì)算值C7H5IN2:243.9497,實(shí)測(cè)值:243.9499)。
