步驟1:合成8-甲基-1,4-二氧雜螺[4.5]癸烷-8-醇:在氬氣保護(hù)下,將1,4-二氧雜螺[4.5]癸烷-8-酮(7.8g,50.0mmol)溶于干燥的THF(75mL)中,冷卻至-78℃。緩慢加入甲基鋰溶液(1.5M在乙醚中,43.3mL,65.0mmol),控制反應(yīng)溫度不超過-55℃。加畢,將反應(yīng)混合物在-55℃下繼續(xù)攪拌4小時(shí)。隨后,將反應(yīng)體系緩慢升溫至室溫,并用飽和NH4Cl溶液淬滅反應(yīng)。分離有機(jī)層,真空濃縮,所得殘余物通過硅膠柱色譜法(洗脫劑比例:乙酸乙酯:石油醚 = 1:9至1:3)純化,得到目標(biāo)產(chǎn)物8-甲基-1,4-二氧雜螺[4.5]癸烷-8-醇,為白色固體(6.8g,收率:80%)。
