以溴化芐和丁香醛為原料合成4-芐氧基-3,5-二甲氧基苯甲醛(5'-1)的一般步驟:將丁香醛(6.0g,32.9mmol)和芐基溴(5.9mL,49.4mmol)溶解于70mL四氫呋喃(THF)中,分批加入至含有氫氧化鉀(3.69g,65.8mmol)的40mL水溶液中。隨后加入催化量的四丁基硫酸氫銨(Bu4NHSO4,0.56g,1.6mmol),反應(yīng)混合物在室溫下攪拌過夜。反應(yīng)完成后,用二氯甲烷(CH2Cl2)萃取,合并有機(jī)相,用無水硫酸鎂(MgSO4)干燥,過濾后減壓濃縮。通過硅膠柱色譜(環(huán)己烷/乙酸乙酯,85:15)純化,得到白色固體狀的4-芐氧基-3,5-二甲氧基苯甲醛(5'-1,8.43g,31.0mmol,產(chǎn)率94%)?;衔锓肿邮綖镃16H16O4,分子量為272.3g·mol?1。分析數(shù)據(jù)與文獻(xiàn)報(bào)道一致(Bennett C.J., Caldwell S.T., McPhail D.B., Morrice P.C., Duthie G.G., Hartley R.C., Bioorg.
