以1-(4-氟苯基)-1-乙醇為原料合成1-(1-溴乙基)-4-氟苯的一般步驟:將1-(4-氟苯基)乙醇(0.455 mL,3.57 mmol)與三溴化磷(0.673 mL,7.13 mmol)在氯仿(10 mL)中混合,于70℃下加熱反應(yīng)3天。反應(yīng)完成后,將反應(yīng)混合物倒入冰水中淬滅,并用乙酸乙酯稀釋。分離有機層,依次用水和鹽水洗滌,無水硫酸鈉干燥后,真空濃縮得到粗產(chǎn)物。粗產(chǎn)物通過硅膠柱色譜法,以己烷/二氯甲烷(2:1)為洗脫劑進行純化,得到1-(1-溴乙基)-4-氟苯(154 mg,0.758 mmol,產(chǎn)率21.3%),為無色油狀物。1H NMR(400 MHz,CDCl3)δ 7.49-7.40(m,2H),7.09-7.00(m,2H),5.23(q,J = 7.0 Hz,1H),2.06(d,J = 6.8 Hz,3H)。
