以甲醇和DL-焦谷氨酸為原料合成(S)-(+)-2-吡咯烷酮-5-羧酸甲酯的一般步驟如下:將DL-焦谷氨酸(2150 g,16.65 mol)溶解于甲醇(2.0 L)和氯仿(2.0 L)的混合溶劑中,加入甲磺酸(80 g,0.83 mol)。將此混合物轉(zhuǎn)移至裝有3?分子篩的索氏提取裝置中,在機(jī)械攪拌下回流反應(yīng)5天。反應(yīng)完成后,通過減壓蒸餾除去溶劑,得到(S)-(+)-2-吡咯烷酮-5-羧酸甲酯,為黃色油狀物,產(chǎn)率為98%(16.3 mmol)。產(chǎn)物無需進(jìn)一步純化即可用于下一步反應(yīng),其物理化學(xué)性質(zhì)與文獻(xiàn)報(bào)道一致。1H NMR(CDCl3, 400 MHz)δ ppm:2.08-2.50(m, 4H, CHCH2CH2),3.77(s, 3H, CH3),4.29(m, 1H, CH2CH2CH),7.62(br s, 1H, NH)。
