將12g(73.1mmol)甲基 ALPHA-D-呋喃木糖苷置于250mL茄形燒瓶中,加入160mL分析純的DMF,冰浴冷卻下分批加入17g NaH(60%分散于礦物油中)。隨后,緩慢滴加38mL溴化芐,滴加完畢后,逐漸升溫至室溫并攪拌過夜。反應(yīng)進(jìn)程通過TLC監(jiān)控。反應(yīng)完成后,小心加入甲醇淬滅反應(yīng),然后用乙酸乙酯(EA)萃取反應(yīng)混合物。有機(jī)相用水洗滌三次,無水硫酸鈉干燥,過濾并濃縮。粗產(chǎn)物通過柱色譜純化(洗脫劑:石油醚/乙酸乙酯=15:1至9:1),得到目標(biāo)化合物(2R,4S)-3,4-雙(芐氧基)-2-((芐氧基)甲基)-5-甲氧基四氫呋喃(28.6g,65.8mmol,收率90%)。
