以4-乙基苯酚為原料合成2-溴-4-乙基苯酚的一般步驟如下:在0℃下,將溴(11.6 mL,0.23 mol)緩慢滴加到冷卻的4-乙基苯酚(25 g,0.21 mol)的二氯甲烷(125 mL)溶液中。滴加完畢后,將反應混合物繼續(xù)攪拌5分鐘,隨后用1N NaOH溶液淬滅反應。將反應混合物用水稀釋后,分離有機層和水層。有機層經濃縮得到橙色油狀物。通過快速柱色譜法(洗脫劑:己烷/乙酸乙酯,0至5%梯度)純化,得到透明油狀的2-溴-4-乙基苯酚(42 g,收率98%)。產物經1H NMR(400 MHz,CDCl3)表征:δ 1.20(t,J = 7.6 Hz,3H),2.58(q,J = 7.5 Hz,2H),5.36(s,1H),6.93(d,J = 8.3 Hz,1H),7.03(d,J = 8.3 Hz,1H),7.28(s,1H)。
