以4-羥基-5-碘嘧啶為原料合成4-氯-5-碘嘧啶的一般步驟如下:在0℃下,向含有7.7 mL(99 mmol)N,N-二甲基甲酰胺(DMF)和150 mL二氯乙烷的攪拌溶液中緩慢滴加12.7 mL(144.6 mmol)草酰氯,以控制劇烈的氣體逸出。待氣體停止釋放后,加入10.0 g 4-羥基-5-碘嘧啶,將反應(yīng)混合物加熱回流3小時(shí)。反應(yīng)完成后,冷卻至室溫,將反應(yīng)液在水和二氯甲烷之間分配。分離有機(jī)層,用無水硫酸鎂(MgSO?)干燥,減壓濃縮除去溶劑,得到9.6 g(收率88%)目標(biāo)產(chǎn)物4-氯-5-碘嘧啶。產(chǎn)物經(jīng)1H-NMR(300 MHz, CDCl?)確認(rèn):δ 8.89(s, 1H)和8.98(s, 1H);電噴霧質(zhì)譜(ESIMS)顯示:[M + H]+ m/z 241.1。
