制備方法

步驟2:4,6-二氯-5-硝基煙酸乙酯(15)的制備 向冷卻至0℃的4,6-二羥基煙酸乙酯(14)(40.0g,0.22mol)的濃硫酸(330ml)溶液中,緩慢滴加發(fā)煙硝酸(12.7ml)。滴加完畢后,將反應(yīng)混合物于0℃下攪拌1小時(shí),隨后升至室溫繼續(xù)攪拌1小時(shí)。反應(yīng)完成后,將混合物緩慢倒入冰水中,劇烈攪拌10分鐘。通過(guò)過(guò)濾收集析出的沉淀,用水充分洗滌,并于真空下干燥,得到淺黃色固體產(chǎn)物15(38.7g,產(chǎn)率77.6%)。產(chǎn)物熔點(diǎn)為238-240℃。 1H NMR(300MHz,DMSO-d6)δ(ppm):8.22(s,1H),4.31(q,J = 7.0Hz,2H),1.29(t,J = 7.0Hz,3H)。 MS(ESI(+)70V)m/z:227 [M-H]-。
