向在-78℃、N2氣氛下保持的1-溴萘(12, 41.41 g, 0.2 mol)的干燥THF(400 mL)攪拌溶液中,通過注射器緩慢滴加1.6 M n-BuLi的正己烷溶液(125 mL, 0.2 mol)。滴加完畢后,將所得溶液在-78℃下繼續(xù)攪拌1小時(shí)。隨后,通過注射器向反應(yīng)體系中滴加干燥的丙酮(17.42 g, 0.3 mol)。反應(yīng)混合物在-78℃下攪拌0.5小時(shí)后,升至室溫并繼續(xù)攪拌1小時(shí)。將反應(yīng)混合物緩慢倒入冰水(2 L)中,攪拌后用CH2Cl2(500 mL × 3)進(jìn)行萃取。合并有機(jī)相,用5%鹽水(500 mL)洗滌,無水Na2SO4干燥,減壓濃縮得到粗產(chǎn)物。粗產(chǎn)物通過柱色譜純化,再用正己烷/EtOAc(1:100, v/v)研磨,得到純品2-(萘-1-基)-2-丙醇(13),為無色針狀晶體(33.90 g, 91%)。熔點(diǎn):88.5-90°C(文獻(xiàn)值:83-85°C [18])。1H NMR (DMSO-d6, 400 MHz) δ: 8.84-8.87 (m, 1H), 7.87-7.90 (m, 1H), 7.78 (d, 1H, J = 8.0 Hz), 7.56 (dd, 1H, J = 0.8, 7.6 Hz), 7.43-7.49 (m, 2H), 7.40 (t, 1H, J = 7.8 Hz), 5.24 (s, 1H), 1.70 (s, 6H)。
