以尿素和2-氨基-4-甲氧基苯甲酸為原料合成7-甲氧基-2,4(1H,3H)-喹唑啉二酮的一般步驟如下:將2-氨基-4-甲氧基苯甲酸(1g,5.98mmol)與尿素(7.18g,120mmol)混合,在155℃下攪拌反應(yīng)16小時。反應(yīng)完成后,將混合物冷卻至100℃,加入3mL水。繼續(xù)冷卻至室溫后進行過濾。向濾液中加入30mL 1mol/L NaOH水溶液以溶解沉淀。1小時后,緩慢滴加4.2mL乙酸,生成淺棕色沉淀,經(jīng)過濾并干燥,得到目標(biāo)產(chǎn)物7-甲氧基-2,4(1H,3H)-喹唑啉二酮(0.55g,2.9mmol,收率48%)。產(chǎn)物表征數(shù)據(jù)如下:1H NMR (500 MHz, DMSO) δ 11.10 (brs, 1H, NH), 10.55 (brs, 1H, NH), 7.8 (d, J = 8.63 Hz, 1H, Ar-H), 6.77 (dd, J = 2.44, 8.84 Hz, 1H, Ar-H), 6.64 (d, J = 2.43 Hz, 1H, Ar-H), 3.82 (s, 3H, OCH3)。13C NMR (125 MHz, DMSO) δ 164.8, 162.8, 151.0, 143.3, 129.3, 111.0, 108.2, 98.8, 56.1。HRMS-ESI (m/z) 計算值 C9H8N2O3 [M + H]+: 193.0489,實測值: 193.0511。
