以5-苯基惡唑?yàn)樵虾铣?-氯-5-苯基惡唑的一般步驟:向圓底燒瓶中加入5-苯基惡唑(1.0 g,6.89 mmol)和無水THF(10 mL)。將反應(yīng)體系冷卻至-78℃。在-78℃下,緩慢滴加2.5 M正丁基鋰(nBuLi,3.03 mL,7.58 mmol)至反應(yīng)液中,反應(yīng)液變?yōu)樯罴t色。保持-78℃反應(yīng)15分鐘后,加入六氯乙烷(1.170 mL,10.33 mmol)。隨后,在2小時(shí)內(nèi)將反應(yīng)體系緩慢升溫至室溫,然后將反應(yīng)液倒入冰水中淬滅。用乙酸乙酯(EtOAc,2×30 mL)萃取水相。合并有機(jī)相,用飽和氯化鈉(NaCl)水溶液洗滌,經(jīng)無水硫酸鎂(MgSO4)干燥,過濾并減壓濃縮。通過硅膠柱色譜(ISCO系統(tǒng))純化粗產(chǎn)物,采用0-100%乙酸乙酯/正己烷梯度洗脫,得到目標(biāo)產(chǎn)物2-氯-5-苯基惡唑(600 mg,3.34 mmol,49%產(chǎn)率),為無色液體。產(chǎn)物經(jīng)質(zhì)譜分析(計(jì)算值:C9H6ClNO m/z 179.1,實(shí)測值:180.1 [M+H]+);1H NMR(500 MHz,CDCl3)δ 7.60(d,J=7.15 Hz,2H),7.43(t,J=7.42 Hz,2H),7.38-7.33(m,1H),7.29(s,1H);13C NMR(126 MHz,CDCl3)δ 153.78,146.16,129.01,126.93,124.03,123.31。
