制備5-溴-2-氯-4-(甲硫基)嘧啶的一般步驟:將2g甲硫醇鈉(28.5mmol,1當(dāng)量)和6.5g 5-溴-2,4-二氯嘧啶(28.5mmol,1當(dāng)量)在50mL無(wú)水乙腈中于室溫下攪拌反應(yīng)24小時(shí)。反應(yīng)完成后,將混合物倒入水中,用二氯甲烷萃取,有機(jī)相用無(wú)水硫酸鈉干燥,隨后減壓濃縮除去溶劑。粗產(chǎn)物通過(guò)己烷重結(jié)晶純化,得到4g目標(biāo)化合物(產(chǎn)率70%)。1H-NMR(400MHz,CDCl3)數(shù)據(jù):δ 8.31(s,1H),2.59(s,3H)。
