以乙酐和3-溴-1-丙醇為原料合成3-溴乙酸丙酯的一般步驟:向攪拌的3-溴-1-丙醇(8.0 g,57.55 mmol)的無水二氯甲烷(40 mL)溶液中加入三乙胺(12 mL,86.32 mmol),并在室溫下攪拌1小時。在0℃下,逐滴加入溶于二氯甲烷(5 mL)的乙酐(6.5 mL,69.06 mmol),隨后在室溫下攪拌16小時。反應(yīng)進程通過薄層色譜(TLC)監(jiān)測。反應(yīng)完成后,用1N鹽酸溶液淬滅反應(yīng)混合物,并用二氯甲烷萃取。合并的有機層用鹽水洗滌,無水硫酸鈉干燥,減壓濃縮。殘余物通過柱色譜法純化,洗脫劑為10%乙酸乙酯/己烷,得到目標產(chǎn)物3-溴乙酸丙酯。產(chǎn)量:10.0 g,產(chǎn)率96%;1H NMR(400 MHz,CDCl3)δ 4.19-4.17(m,2H),3.47(t,J=6.5 Hz,2H),2.21-2.18(m,2H),2.06(s,3H)。
