實(shí)施例1c:2-(甲硫基)-4-氯-6-甲基嘧啶-5-甲酸乙酯的合成 在100 mL圓底燒瓶中加入2-(甲硫基)-4-甲基-6-氧代-1,6-二氫嘧啶-5-甲酸乙酯(6.49 g,28.4 mmol)和14 mL三氯氧磷(POCl3)。向該溶液中加入N,N-二異丙基乙胺(1.2 mL),將反應(yīng)混合物在80~90℃下攪拌2小時(shí)。反應(yīng)完成后,將混合物減壓濃縮,得到深棕色殘余物。將殘余物用冰水淬滅,然后用二氯甲烷(DCM)和飽和碳酸氫鈉(NaHCO3)水溶液稀釋。分離有機(jī)層,并用無水硫酸鎂(MgSO4)干燥。通過旋轉(zhuǎn)蒸發(fā)儀除去溶劑,得到深棕色油狀產(chǎn)物2-(甲硫基)-4-氯-6-甲基嘧啶-5-甲酸乙酯(6.76 g,收率96%)。質(zhì)譜(MS)m/z:246.9([M + H]+),248.9(同位素峰)。
