以3-氯-5-硝基異喹啉為原料合成3-氯-5-氨基異喹啉的一般步驟:將3-氯-5-硝基異喹啉(2.3g,11.0mmol)溶解于乙酸(100mL)中,隨后分批加入鋅粉(3.5g,55.1mmol)。將反應(yīng)混合物在室溫下攪拌2小時(shí)。反應(yīng)完成后,通過硅藻土過濾混合物,濾液在減壓條件下濃縮。殘余物通過硅膠柱色譜法進(jìn)行純化,使用含1%甲醇的二氯甲烷溶液作為洗脫劑,最終得到3-氯-5-氨基異喹啉,為灰白色固體。產(chǎn)物經(jīng)質(zhì)譜(ESI,m/z)分析顯示:[M+1]+峰位于179.0;1H NMR(300MHz,DMSO-d6)δ值分別為8.98(s,1H),8.13(s,1H),7.40-7.35(m,1H),7.28-7.25(m,1H),6.90-6.87(m,1H),6.06。
