以1-甲基-4-(6-硝基吡啶-3-基)哌嗪為原料合成1-甲基-4-(6-氨基吡啶-3-基)哌嗪的一般步驟:將1-甲基-4-(6-硝基吡啶-3-基)哌嗪(1.11 g,5.02 mmol)和10% Pd/C(110 mg)在甲醇(15 mL)中的混合物于室溫下攪拌5小時。反應(yīng)完成后,通過過濾移除催化劑,濃縮濾液,得到951 mg(收率99%)的黃色固體產(chǎn)物1-甲基-4-(6-氨基吡啶-3-基)哌嗪,該產(chǎn)物無需進一步純化即可用于后續(xù)反應(yīng)。產(chǎn)物的1H NMR(CDCl3)數(shù)據(jù)如下:δ 7.78(d,1H,J = 3.0 Hz),7.18(dd,1H,J = 3.0, 8.9 Hz),6.48(d,1H,J = 8.9 Hz),3.08(dd,4H,J = 4.9, 4.9 Hz),2.61(dd,4H,J = 4.9, 4.9 Hz),2.37(s,3H)。
