以氯乙酰胺和亞磷酸三乙酯為原料合成氨基甲酰甲基膦酸二乙酯的一般步驟:將2-氯乙酰胺(5.01 g,53.6 mmol)和亞磷酸三乙酯(9.19 mL,53.6 mmol)溶于鄰二甲苯(14 mL)中,加熱回流3.5小時。反應完成后,減壓蒸餾除去溶劑,得到深棕色焦油狀殘余物。將殘余物溶解于二氯甲烷中,通過短硅膠柱過濾。濃縮濾液,所得固體用乙酸乙酯/戊烷混合溶劑重結晶,得到氨基甲酰甲基膦酸二乙酯(3.42 g,產率33%),為淺棕色晶體。產物結構經1H NMR(200 MHz,DMSO-d6)δ 7.35(br s,1H),7.02(br s,1H),4.02(dq,4H,J = 7.1 Hz,JPH = 1.1 Hz),2.80(d,2H,JPH = 21.4 Hz),1.23(t,6H,J = 7.0 Hz);13C NMR(50 MHz,DMSO-d6)δ 166.0(d,2JCP = 5.1 Hz),61.5(d,2JCP = 6.0 Hz),34.5(d,1JCP = 131.6 Hz),16.2(d,3JCP = 6.0 Hz);31P NMR(81 MHz,DMSO-d6)δ 23.8確認。質譜分析結果:ESIMS m/z 218 [M + Na]+;高分辨質譜(HRMS)計算值C6H14NO4P [M]+ 195.0655,實測值195.0653。
