中間體35的制備[通用程序14]: 在干燥條件下,將中間體34(3g,19.6mmol)和乙腈(0.8mL,19.6mmol,1當(dāng)量)溶于甲苯(10mL)中。將此溶液緩慢加入至預(yù)先在甲苯(50mL)中懸浮的氫化鈉(784mg,19.6mmol,1當(dāng)量,60%礦物油分散體)混合物中,反應(yīng)溫度維持在65℃。反應(yīng)混合物在65℃下持續(xù)攪拌16小時(shí)。反應(yīng)完成后,將混合物冷卻至室溫,并用冰冷的水(20mL)淬滅反應(yīng)。通過過濾收集生成的固體,得到中間體35(1.5g,產(chǎn)率53%),為棕色固體。產(chǎn)物經(jīng)1H NMR(CDCl3)鑒定: δ 8.70(d, J = 4.8Hz, 1H), 8.12(d, J = 7.5Hz, 1H), 7.90-7.94(m, 1H), 7.56-7.60(m, 1H), 4.41(s, 2H)。質(zhì)譜分析顯示分子離子峰[M + H]+為147。薄層色譜(TLC)分析,展開劑為乙酸乙酯,Rf值為0.40。
