以6-喹啉羧酸甲酯為原料合成喹啉-6-甲酰肼的一般步驟:將6-喹啉羧酸甲酯(0.10 mol)溶于乙醇(50 mL)中,加入99%水合肼(0.10 mol),將混合物回流17小時。反應(yīng)完成后,冷卻反應(yīng)混合物,過濾收集所得固體,并用少量冷甲醇洗滌,得到喹啉-6-甲酰肼。反應(yīng)進(jìn)程通過薄層色譜法(TLC)監(jiān)測,展開劑為甲苯:丙酮(8:2,v/v)。產(chǎn)物為無色固體,收率89%,熔點195-197℃。IR(KBr,cm^-1):3376, 3291(NH2),1689(C=O,酰胺)。1H NMR(400 MHz,DMSO-d6):δ 10.34(br s,1H,NH),8.89(dd,J = 1.7, 4.1 Hz,1H),8.38-8.43(m,2H),8.19(dd,J = 1.6, 8.3 Hz,1H),7.97(d,J = 7.7 Hz,1H),7.53(dd,J = 4.1, 8.3 Hz,1H),4.71(br s,2H,NH2)。
