以4,6-二氯-5-氨基嘧啶和一甲胺為原料合成6-氯-4-甲基嘧啶-4,5-二胺的一般步驟:將5-氨基-4,6-二氯嘧啶(5,1.0mmol)、三乙胺(1.0mmol)、乙醇(4.3mL)和一甲胺(2.5mmol)的混合物置于鋼瓶中,加熱至55℃反應(yīng)3小時。反應(yīng)完成后,在減壓條件下除去揮發(fā)性物質(zhì)。粗產(chǎn)物通過硅膠柱色譜純化,洗脫劑為CHCl3-cC6H12-CH3OH(70:20:10),隨后用EtOAc重結(jié)晶,得到目標(biāo)化合物6-氯-4-甲基嘧啶-4,5-二胺(6),為白色固體(148mg,收率93%)。產(chǎn)物表征數(shù)據(jù)如下:熔點(diǎn)165-67℃;1H NMR(400MHz,[D6]DMSO)δ 2.89(d,J=4.4Hz,3H,CH3),4.96(br s,2H,NH2),6.86(br q,J=4.4Hz,1H,NH),7.75(s,1H,H-2);元素分析(C5H7ClN4)符合理論值。
