步驟1:4-氟-3-碘-1H-吲唑(B-2)的制備。在室溫下,向攪拌中的4-氟-1H-吲唑(B-i)(5.00 g,36.73 mmol)的DMF(80 mL)溶液中依次加入碘(I2)(18.64 g,73.46 mmol)和氫氧化鉀(KOH)(7.73 g,137.7 mmol)。反應(yīng)2小時(shí)后,薄層色譜(TLC)監(jiān)測(cè)顯示反應(yīng)完成。將反應(yīng)混合物緩慢倒入10%的亞硫酸氫鈉(NaHSO3)水溶液(200 mL)中,并用乙酸乙酯(EA)(3×200 mL)萃取。合并有機(jī)層,依次用水(100 mL)和飽和食鹽水(2×200 mL)洗滌,經(jīng)無(wú)水硫酸鈉(Na2SO4)干燥,過(guò)濾后濃縮。粗產(chǎn)物用石油醚(PE)洗滌,得到黃色固體B-2(8.33 g),產(chǎn)率為86.5%。B-2的物理表征數(shù)據(jù)如下:液相色譜-質(zhì)譜聯(lián)用(LCMS)(電噴霧離子化,ESI):計(jì)算值C7H4FIN2,261.9;實(shí)測(cè)值[M + H]+=262.9。
