以2-溴-4-氟-6-硝基甲苯為原料合成2-甲基-3-溴-5-氟苯胺的一般步驟:[00402]將氯化銨(571 mg,10.7 mmol)加入到1-溴-5-氟-2-甲基-3-硝基苯(500 mg,2.14 mmol)和鐵粉(0.6 g,10.68 mmol)的混合物中。隨后加入水(24 mL)和乙醇(24 mL)。將反應(yīng)混合物回流5小時,反應(yīng)完成后,通過硅藻土過濾,濾餅用乙醇(100 mL)和乙酸乙酯(100 mL)洗滌。真空除去有機(jī)溶劑,得到水溶液,用二氯甲烷(3×30 mL)萃取。分離有機(jī)層并濃縮,得到粗產(chǎn)物3-溴-5-氟-2-甲基苯胺(390 mg,1.87 mmol,產(chǎn)率87.7%),該產(chǎn)物經(jīng)核磁共振(NMR)分析顯示純度良好,無需進(jìn)一步純化即可用于下一步反應(yīng)。[00403] 1H NMR(DMSO-d6):δ 6.60(dd,J = 2.8和8.4 Hz,1H),6.42(dd,J = 2.8和11.6 Hz,1H),5.54(bs,2H),2.09(s,3H)。
