以丙酮酸和4-氨基-3-碘三氟甲苯為原料合成5-三氟甲基吲哚-2-羧酸的一般步驟:將2.28 g 2-碘-4-三氟甲基苯胺、2.10 g丙酮酸、2.67 g DAI3CO、89 mg乙酸鈀(II)和25 mL DMF的混合物在100°C下攪拌24小時(shí),反應(yīng)在氮?dú)夥諊羞M(jìn)行。反應(yīng)完成后,將反應(yīng)混合物冷卻至室溫,加入25 mL乙酸乙酯和50 mL 5M鹽酸,用乙酸乙酯萃取3次。合并有機(jī)層,依次用水和飽和食鹽水洗滌,用無水硫酸鎂干燥,減壓濃縮除去溶劑。殘余物用氯仿洗滌,得到1.15 g 5-三氟甲基吲哚-2-羧酸(以下簡稱“化合物7”)。1H-NMR (DMSO-d6) δ: 13.24 (br s, 1H), 12.20 (br s, 1H), 8.08 (s, 1H), 7.60 (d, 1H, J = 8.8 Hz), 7.51 (dd, 1H, J = 8.8, 1.6 Hz), 7.24 (d, 1H, J = 1.6 Hz)。
