在機械攪拌下,將5-硝基-1-苯并噻吩-2-羧酸(40 g, 179 mmol)溶解于喹啉(350 mL)中,隨后加入銅粉(11.3 g, 179 mmol)。將反應(yīng)混合物加熱至190°C,直至氣體釋放完全。反應(yīng)完成后,將混合物冷卻至室溫,并倒入碎冰中,用濃鹽酸酸化。所得懸浮液用乙酸乙酯萃取,分離有機層,依次用2N鹽酸、水和飽和鹽水洗滌。有機層經(jīng)無水硫酸鈉干燥后,濃縮,得到5-硝基苯并噻吩(30.5 g, 95%收率)。產(chǎn)物結(jié)構(gòu)通過1H NMR(DMSO-d6, 400 MHz)和LC-MS(電噴霧)確認:1H NMR δ 8.85 (d, 1H), 8.31 (d, 1H), 8.18 (dd, 1H), 8.06 (d, 1H), 7.73 (d, 1H); LC-MS m/z = 180 [M + H]+。
