以3-溴-5-甲氧基吡啶(2g,10.63mmol)、雙(頻哪醇合)二硼(4.05g,15.98mmol)和乙酸鉀(4.78g,42.55mmol)為原料,在無水甲苯(25mL)中進(jìn)行反應(yīng)。首先對反應(yīng)體系進(jìn)行氮?dú)饷摎馓幚?。在氮?dú)獗Wo(hù)下,向反應(yīng)體系中加入Pd(dppf)Cl2·DCM(0.87g,0.10mmol),隨后將反應(yīng)混合物加熱回流2小時(shí)。反應(yīng)進(jìn)程通過薄層色譜(TLC)監(jiān)測。反應(yīng)完成后,將混合物冷卻至室溫(25℃),通過Celite墊過濾。濾液用乙酸乙酯(100mL)稀釋,依次用水和鹽水洗滌,無水硫酸鈉干燥后減壓濃縮,得到粗產(chǎn)物。粗產(chǎn)物通過硅膠(100-200目)柱色譜法純化,洗脫劑為10%乙酸乙酯的己烷溶液,最終得到2.6g目標(biāo)產(chǎn)物5-甲氧基-3-吡啶頻哪醇硼酸酯。經(jīng)LC-MS分析,產(chǎn)物分子量為236(M+H)。
