以2,7-二溴咔唑和碘苯為原料合成2,7-二溴-9-苯基-9H-咔唑的一般步驟:將32.5g(100毫摩爾)2,7-二溴-9H-咔唑、20.4g(100毫摩爾)碘苯、9.5g(150毫摩爾)銅粉、27.6g(200毫摩爾)碳酸鉀和600ml二甲基甲酰胺混合,在氮氣保護(hù)下于130℃加熱反應(yīng)過夜。反應(yīng)完成后,冷卻至室溫,過濾反應(yīng)混合物。濾液用二氯甲烷和水進(jìn)行三次萃取,有機(jī)相用無水硫酸鎂干燥,隨后在真空下蒸發(fā)除去溶劑。殘余物通過硅膠柱色譜法(洗脫劑:己烷-二氯甲烷)純化,得到白色固體產(chǎn)物2,7-二溴-9-苯基-9H-咔唑(31.3g,78毫摩爾,收率78%)。
