以2-氟-5-溴吡啶和聯(lián)硼酸頻那醇酯為原料合成2-氟吡啶-5-硼酸頻哪醇酯的一般步驟:在氮?dú)獗Wo(hù)下,將Pd(dppf)Cl2(4.16g,5.68mmol)一次性加入到含有5-溴-2-氟吡啶(10g,56.82mmol)、聯(lián)硼酸頻那醇酯(21.64g,85.23mmol)和乙酸鉀(16.73g,170.46mmol)的二惡烷(100mL)溶液中?;旌衔镌谑覝叵聰嚢?0分鐘后,加熱回流6小時(shí)。反應(yīng)完成后,直接蒸發(fā)去除溶劑,殘余物通過閃式硅膠柱色譜法純化,得到目標(biāo)產(chǎn)物2-氟吡啶-5-硼酸頻哪醇酯(10.5g,產(chǎn)率82.85%)。產(chǎn)物的結(jié)構(gòu)通過1H NMR(400MHz,氯仿-d)確認(rèn):δ 8.59(s,1H),8.15(dt,J = 1.76, 8.41 Hz,1H),6.91(dd,J = 2.26, 8.28 Hz,1H),1.35(s,12H)。
