在氮?dú)獗Wo(hù)下,將25%甲醇鈉的甲醇溶液(16 mL)緩慢滴加至攪拌中的2-溴丙酸(19.6 mmol)的甲醇(5 mL)溶液中。反應(yīng)混合物在50℃下加熱反應(yīng)過夜,同時(shí)持續(xù)通入氮?dú)獗Wo(hù)。反應(yīng)完成后,通過旋轉(zhuǎn)蒸發(fā)儀在減壓條件下濃縮反應(yīng)液。殘余物用1N鹽酸水溶液調(diào)節(jié)至pH 1,隨后用乙酸乙酯進(jìn)行三次萃?。?0 mL、25 mL、10 mL)。合并有機(jī)相,用無水硫酸鈉干燥,過濾后再次減壓濃縮,得到2-甲氧基丙酸,為無色油狀物,產(chǎn)量2.04 g(收率99%),其純度可直接用于后續(xù)反應(yīng)無需進(jìn)一步純化。產(chǎn)物經(jīng)1H NMR(CD3OD)表征:δ 3.67(q, 1H),3.33(s, 3H),1.33 ppm(d, 3H)。
