以5-溴-4-甲基-3-硝基吡啶-2-胺(700.0 mg,3.02 mmol)為原料,加入鐵粉(1680.0 mg,30.20 mmol)和濃鹽酸(50.0 μL)于乙醇(2.8 mL)和水(0.7 mL)的混合溶劑中。將反應(yīng)懸浮液在100℃下攪拌30分鐘。反應(yīng)完成后,將混合物冷卻至室溫,通過硅藻土過濾以去除不溶性雜質(zhì)。濾液經(jīng)減壓蒸餾濃縮,所得殘余物通過硅膠柱色譜法(洗脫劑:甲醇/二氯甲烷 = 1:40)進行純化。收集含有目標產(chǎn)物的級分,經(jīng)蒸發(fā)除去溶劑后,得到5-溴-4-甲基吡啶-2,3-二胺的棕色固體(550.0 mg,收率90%)。產(chǎn)物經(jīng)LCMS ESI(+)分析:m/z 202([M + H]+),204([M + H + 2]+)。1H-NMR(300 MHz,DMSO-d6)數(shù)據(jù):δ 7.37(s,1H),5.49(s,2H),4.74(s,2H),2.12(s,3H)。
