以1,2,3,4-四氫喹啉-2-羧酸乙酯為原料合成(S)-(1,2,3,4-四氫喹啉-2-基)甲醇的一般步驟如下:在0℃條件下,將1,2,3,4-四氫喹啉-2-羧酸乙酯(25mmol)的四氫呋喃溶液(5ml/mmol)緩慢滴加到氫化鋁鋰(2當量)的四氫呋喃懸浮液(50ml)中。反應混合物在25℃下攪拌1小時后,升溫至回流狀態(tài)繼續(xù)反應4小時。反應進程通過薄層色譜(TLC)進行監(jiān)測。反應完成后,向混合物中緩慢加入飽和硫酸鈉溶液淬滅反應,隨后通過硅藻土過濾,并用乙酸乙酯洗滌濾餅。合并濾液后,在減壓條件下濃縮去除溶劑,所得粗產(chǎn)物通過柱色譜法(洗脫劑比例為3:7的乙酸乙酯/己烷)進行純化。最終產(chǎn)物的收率為50%。
