以2-氯吡啶-5-磺酰氯為原料合成6-氯-3-磺胺吡啶的一般步驟如下:在-50℃條件下,將6-氯吡啶-3-磺酰氯(5.0g,0.024mol)溶解于0.5M氨的二惡烷溶液(125mL)中。將反應(yīng)混合物緩慢升溫至室溫,并持續(xù)攪拌1小時。反應(yīng)完成后,將混合物通過硅藻土過濾,用二惡烷洗滌濾餅兩次。濾液經(jīng)真空濃縮后,得到6-氯吡啶-3-磺酸酰胺,為灰白色固體,產(chǎn)量4.55g(收率98%)。產(chǎn)物經(jīng)液相色譜(LC)分析,檢測條件為UV215nm;保留時間(Rt)1.05分鐘,純度100%。質(zhì)譜(m/z)分析結(jié)果(ES+):193/195。核磁共振氫譜(400MHz; d6-DMSO)數(shù)據(jù):δ8.79(1H,d),8.21(1H,dd),7.75(1H,d),7.70(2H,br s)。
