以1-羥基異喹啉為原料合成4-溴-1(2H)-異喹啉酮的一般步驟:向含有異喹啉-1(2H)-酮(105 mg,0.723 mmol)的DMF(2 mL)溶液中加入N-溴代琥珀酰亞胺(NBS,142 mg,0.796 mmol)。將反應混合物在室溫下攪拌2小時。反應完成后,將混合物濃縮以去除溶劑。粗產(chǎn)物通過制備型高效液相色譜(HPLC)進行純化,得到中間體6 4-溴異喹啉-1(2H)-酮(110 mg,產(chǎn)率68%)。[C] MS(ESI)m/z:223.9(M + H)+。1H NMR(500 MHz,DMSO-d6)δ11.57(br.s,1H),8.24(dd,J = 8.0, 0.8 Hz,1H),7.88-7.83(m,1H),7.79-7.75(m,1H),7.61(ddd,J = 8.0, 7.1, 1.1 Hz,1H),7.55(s,1H)。
